To explain this: Normally the DTR line is pulled down to reset the arduino.
After the reset the arduino starts the bootloader and accepts the serial upload.
If the DTR line is broken you can manually reset the arduino, you just need the right timing.
I can see it in my gateway.
I only send 1 from my node and then use a timer in Domoticz.

I have change my node and you was right. When you send both 1 and 0 then it worked.
With the voltage regulator removed the Arduino can no longer be powered on the raw pin. You need to supply it with maximum 3.3V on Vcc. 2xAA usually works well.
After a few hours of trying this i now working.
I reconnect all the wiring and then it start to work.
It must have been som problem with the wiring.
Now i use Arduino 1.6.5 and develop version of mysensors API.

I had the same issue, pressing reset didn't work but reconnecting the USB cable did the trick.
